What are Oligonucleotides?

Oligonucleotides (ONs) are a class of drugs composed of 12 to 30 single- or double-stranded ribose oligonucleotides synthesized chemically. They act on the target messenger RNA (mRNA) through the Watson-Crick base pairing principle, mainly divided into the following types: antisense oligonucleotides (ASOf) and small interfering RNA (siRNA), micro-RNA, aptamers, and so on. In general, natural oligonucleotides have poor stability and are easily degraded by nucleases in vivo. They have high polarity, low specificity, and poor affinity for target mRNAs. By chemical modification, specific groups are introduced into oligonucleotides, which can effectively enhance the stability of oligonucleotides and reduce toxic and side effects.

Application of Oligonucleotides

Utilizing oligonucleotides as drugs is a completely new therapeutic approach compared to traditional small-molecule drugs. Oligonucleotides are essential tools for regulating gene expression in biomedical and life science research and have been developed as gene-targeted therapeutics for the treatment of viruses, tumors, and genetic diseases. Oligonucleotide drugs such as Givosiran, Lumasiran, and Viltolarsen, which have received F DA approval in recent years, have led the way for RNA interference or RNA therapy to enter the mainstream of drug development.
